Clinical Guidelines: The Scientific Foundation

A common misconception is that precision oncology represents an alternative to guideline-based medicine. In fact, international clinical guidelines remain the essential foundation of every treatment decision, as they systematically summarize the current state of scientific evidence. What Role Do Off-Label Therapies Play?

In precision oncology, a molecular alteration may be identified for which a potentially suitable medication already exists, even though the drug is not formally approved for the patient’s specific tumor type.

When a medication is used outside the indications covered by its official authorization, this is referred to as off-label use.

Modern clinical guidelines increasingly take tumor-agnostic biomarkers into account. If molecular analysis identifies a potential target for which there is credible evidence of clinical benefit, the option of off-label treatment may be evaluated individually following a careful review of the available studies and a thorough assessment of potential benefits and risks.
